چکیده

Novel cationic glycoliposomes, composed of 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC), cholesterol (Chol) and glycoamphiphiles featuring a galactosyl, mannosyl or glucosyl moiety have been investigated for the targeted delivery trans-resveratrol (RSV), Quorum Sensing Inhibitor (QSI), to Methicillin Resistant Staphylococcus Aureus (MRSA) biofilms. All glycosylated formulations show 10–20 % reduction their hydrodynamic size high positive increase in ?-potential (20÷27mV), with respect almost neutral DOPC/Chol liposomes (-3.7 mV). RSV is entrapped Entrapment Efficiency (EE%), containing amphiphiles showing higher values EE% (79–90 %) than those DOPC/CHOL (65 %). In all liposomal formulations, inclusion causes decrease ?-potential, which particularly evident negative value (-14.8 This probably due ionization small percentage molecules that point towards lipid/water interface, as reported literature. Greater antioxidant activity found when embedded liposomes, rather liposomes. finding suggests different distribution lipid membrane enclosing amphiphiles, favor an external exposure RSV. Biological assays carried out monitor demolition effect RSV-loaded on mature biofilm MRSA presence essential take place matrix. particular, RSV-galactosylated are most effective destroying even at concentration (0.019 mM) sixty times lower MIC (1.2 mM). work demonstrates, first time, how functionalization glycosydic residues can enhance liposome performances QSI nanocarriers treatment associated infections.

Efficacy of combination oral antimicrobial agents against biofilm-embedded methicillin-resistant Staphylococcus aureus.

BACKGROUND The combination of fusidic acid and rifampicin has a demonstrated synergistic effect against methicillin-resistant Staphylococcus aureus (MRSA), including planktonic and biofilm-related organisms. However, the in vitro efficacy of other combinations of oral anti-MRSA antibiotics in biofilm models has not been established. METHODS The antibacterial activity of fusidic acid, linezoli...

Comparison of Biofilm Formation between Methicillin-Resistant and Methicillin-Susceptible Isolates of Staphylococcus aureus

Background: The aim of this study was to compare the biofilm formation and the prevalence of biofilm-associated genes between the isolates of methicillin-resistant (MRSA) and methicillin-susceptible (MSSA) Staphylococcus aureus. Methods: In total, 209 S. aureus isolates were collected. The antibiotic susceptibility test was conducted using nine antibiotics according to the guidelines of Clinica...

Ascorbic acid augments colony spreading by reducing biofilm formation of methicillin resistant Staphylococcus aureus

Objective(s):Staphylococcus aureus is a Gram-positive pathogen, well known for its resistance andversatile lifestyle. Under unfavourable conditions, it adapts biofilm mode of growth. For staphylococcal biofilm formation, production of extracellular polymeric substances (EPS) is a pre-requisite, which is regulated by ica operon-encoded enzymes. This study was designed to know the impact of ascor...

Methicillin Resistant Staphylococcus aureus

Etiology Staphylococcus aureus is a Gram positive, coagulase positive coccus in the family Staphylococcaceae. Methicillin-resistant S. aureus (MRSA) strains are resistant to methicillin and essentially all other beta-lactam antibiotics. MRSA isolates are genetically heterogeneous. 1 Some strains, which are called epidemic strains; are more prevalent and can spread within or between hospitals an...

Battling the Methicillin-Resistant Staphylococcus aureus Biofilm Challenge with Vancoplus

Of 55 MRSA, 47 isolates were biofilm producers and 8 isolates were non-biofilm producers. Of 47 biofilm producers, 24 (51.0%), 14 (29.8%) and 9 (19.1%) produced strong (OD570 ≥ 0.5), medium (OD570 ≥ 0.2 to <0.5), weak (OD570 0 to <0.2) biofilm, respectively. Of the 47 isolates, eight determinants (genes) (eno, hla, hlb, clfA, fnaA, icaA, agrII and sar) were found predominantly among 70 to 80% i...
